Mexico. Two federales, Machete and his partner, are on their way to save a girl who was a witness of a drug kingpin, Torrez. Machete's partner is not too eager to save the girl from the drug lord who keeps her in a hotel. The partner tells Machete that they are federales, not sidekicks and that they don't have to save the girl. Machete, however, doesn't pay too much attention to him and drives on. Machete's boss calls them and tells Machete to turn back because it's too dangerous mission. Machete crushes the cell phone in his fist. When Machete and his partner get to the hotel, the partner ends up saying: "Whatever happens, I'm with you... boss". Machete pulls out his machete and responds: "This is the boss". Then he drives into the hotel, to which entry is surrounded by Torrez's men, who kill Machete's partner while Machete gets closer to the building. Machete steps out of the car and kills numerous people in the hotel, chopping off their heads and limbs, and slashing them in half with his blade. He then finds a naked girl in one of the hotel's bedrooms - the girl that he was to save. He carries her out, but he notices a few more Torrez's men coming in. The girl teases him, and then pulls out his machete and stabs him in the leg with it. Then, Machete's boss shows up and it is revealed that he is corrupted by Torrez. After that, Torrez himself shows up and kills the naked girl. Torrez explains to Machete that he owns the police and many other forces not just in Mexico. He then makes his men bring Machete's wife and he cuts her head off while Machete watches in horror. Torrez then implies that Machete's daughter is dead, as well. Machete is desperate to die, but Torrez does not cut his head off. Instead, he walks out and tells his men to burn the hotel down. As they do that, Machete's face is grim and furious.The opening credits roll, presenting the cast and the other makers of the movie. As they are presented, a re-written, instrumental version of Tito & Tarantula's song "Machete" is heard. The music heard in the opening credits is called "Machete Opening Titles" or is also referred to as "Machete Deluxe".
An injured Machete is taken to a hospital to be treated for wounds, and escapes from Booth's henchmen sent to kill him. Agent Yvetta Sartana (Jessica Alba), a persistent U.S. Immigration and Customs Enforcement agent, is sent by her superior to go find and capture the injured Machete. Machete, with the help of Shé (Michelle Rodriguez) recruits Padre (Cheech Marin), Machete's "holy" brother. To take revenge on Booth, Machete kidnaps Booth's wife and his daughter, April (Lindsay Lohan), who has a penchant for guns. After encountering Machete, Sartana begins to develop an interest in him. Machete rallies a group of illegal immigrants together, and begins to hunt the people who double crossed him.
